1. 96 deep well plate is used for growing bacteria cells.

2. Bacteria is usually growing in 1.3-1.5 ml of rich medium such as 2X YT broth + 70-100 ml/ml Ampicillin.

3. Longer time may be required. Incubation time ranges from 18-24 hours.

4. Higher shaking speed may be beneficial (250-350 rpm).
