Estimation of protein concentration --- Bradford (I)

This protocol uses the Bradford Protein assay kit (e.g., BioRad Protein Assay Kit).

1. Add 0.5, 1.0, 2.0, 5.0, 10.0 mg of BSA to tubes containing 800 ml of water. Add 1 and 5 ml of your sample to two tubes containing 800 ml of water.

2. Add 200 ml of the BioRad reagent and mix.

3. Let stand 5 minutes and read absorbance at 595 nm.

4. Make a standard curve using the BSA reading and estimate the protein concentration in the sample.

Note: Bradford assay will not be accurate for samples containing high concentration of detergent.
