1. Dissolve p-phenylenediamine (10 mg) or n-propyl gallate (50 mg) in
1 ml of PBS or 0.1 M Tris-HCl pH 9.0. Keep wrapped in the dark.
2. Add 9 ml of glycerol. Mix by stirring. Keep wrapped in the dark.
3. (optional) Adjust pH to ≥ 8.0 using 0.1 M sodium carbonate buffer, pH 9.2.
(mix 0.2 M sodium carbonate and 0.2 M sodium bicarbonate 1:11.5)
4. Store at -20 °C in the dark.
Other antiquench agents:
1,4-Diazabicyclo (2,2,2)-octane (DABCO) (~2%)
Ascorbic acid (vitamin C)
Mowiol (10%) or Gelvatol (15%)
5. p-phenylenediamine may cause Cy-dye to fade, use n-propyl gallate
instead.
