Sf9 and Sf21 cells can be grown as monolayer cultures using flasks
or tissue culture plates. Cells can be detached by pipetting the medium
across the monolayer. No Trypsin-EDTA is necessary. These cells can be grown
at room temperature or in a 27 °C incubator. They do not require CO2
incubator.
Sf9 and Sf21 cells can also be grown in suspension in a shaker or spinner.
Seed cells at 0.5-1x 106 cells/ml in a shaker
or a roller bottle. Cells transfered from monolayer to suspension may not
grow well initially. Speed of shaking or spinning should increase gradually
to adapt cells in the suspension culture.
